These are useful internet resources that our laboratory uses on a daily
basis. We use them for design and ordering of primers for PCR and PCR cloning,
reading of our DNA sequencing reactions (when we are using dye-tagged),
finding gene structures in human and mouse genome data that match our cDNAs
etc. Our Yeast Two Hybrid protocols for transformation are based on Dan
Gietz's lab protocols at the University of Manatoba (see list below). We
typically get 1-5 million transformants per screen from 10-2- ng of library
plasmid DNA. Thanks to Dan for advice in transforming yeast when I was
a humble post doc at the Kellogg Eye Center (U. Mich.) starting off with
yeast two-hyrbid methods for the first time.
